SOP on sampling and testing procedure of swab sample

1.0       OBJECTIVE:

            To lay down a procedure for sampling and testing procedure of swab sample.

2.0       SCOPE:

This Standard Operating Procedure is applicable for sampling and testing procedure of swab sample in Microbiology Laboratory.

3.0       RESPONSIBILITY:

            Microbiologist shall be responsible to follow the procedure mention in this SOP.

4.0       ACCOUNTABILITY:

Department Head & QA Head shall be accountable for effective implementation of this SOP.

5.0       ATTACHMENTS:

Report for Total Microbial Count of swab sample                   – Attachment I

6.0       PROCEDURE :

6.1       Prepare the sterile swab in 0.9% saline (10ml) solution in the test tube having cotton plug or use readymade sterile moisten swab (Himedia).

6.2       The swabbing shall be done on the area of 100cm2 by following horizontal strokes first followed by vertical strokes.

6.3       Place the swab back in the test tube and vortex. Incase  of readymade sterile moisten

Swab pour about 10 ml of 0.9% saline in tube and vortex.

6.4       Filter the content through 0.45m membrane filter and place the filter on the Petri plate

having sterile SCDA medium.

6.5       Incubate the plates at 30- 35°C for 48 hrs and further at 20-25°C for 72 hrs.

6.6       Count the colonies and report the total microbial count as CFU/100cm2 in Attachment –I .
